Abstract

Alterations of the endolysosomal and autophagic pathways have been associated with several disorders, including some neurological conditions. Accordingly, defects in the endo-lysosomal pathway and in autophagy/mitophagy pathway have been observed in Alzheimer’s disease, ischemic stroke and Amyotrophic Lateral Sclerosis.
Early pathological changes in both systems compromise the removal of misfolding & aggregating proteins as well as increase damaged mitochondria, leading to a gradual accumulation of defective autophagic vacuoles (AVs) and lysosome dyshomeostasis.
Relevant therapeutic strategies aiming at restoring the homoeostasis of these pathways can foster neuronal survival.
