Fibrodysplasia Ossificans Progressiva (FOP) is a rare congenital disorder causing abnormal bone growth in muscles due to a gene mutation (R206H) in Acvr1, activating bone morphogenetic proteins and activin A pathways. Inflammatory flare-ups trigger this process. Macrophages' role in affected tissues is unclear. Using mouse models, we simulated acquired and genetic HO, investigating the innate immune system's role in bone formation. Single-cell transcriptomics and imaging suggest early collaboration between macrophages and mesenchymal cells, fostering bone formation.
